Personally, I think from experience and looking at google results, and also from a user point of view domain.com/keyword is the best way to go.

If you used www.keyword.mydomain.com you would then presumably have www.keyword.mydomain.com/keyword/ anyway? Which would possibly tag you as spamming the URL for SEP purposes.
In any event you would have keyword.mydomain.com not www.keyword.mydomain.com
In the end the difference to the SEO is nothing, or near as damn nothing, whereas the difference to your visitors is considerable - so forget SEs in the desician and go with www.mydomain.com/keyword
My understanding is that www.mydomain.com/keyword is better and would be considered part of your domain and if you go the other way, your primary domain will not pass on any PR to your sub domain.
